As a trusted peptide vendor uk and leading EU peptide supplier, we provide researchers across Europe with this complete research kit for sale that delivers exceptional purity, consistency, and documented quality for studies in melanogenesis, receptor pharmacology, and metabolic regulation .

Melanotan II (MT-2) is a synthetic cyclic heptapeptide analog of alpha-melanocyte-stimulating hormone (α-MSH), originally developed at the University of Arizona in the 1980s to increase melanin formation and protect the skin from harmful UV radiation . The peptide has the cyclic structure Ac-Nle4-Asp5-His6-D-Phe7-Arg8-Trp9-Lys10 α-MSH4-10-NH2, with the crucial D-Phe-7 substitution that contributes to its superagonist properties and enhanced potency compared to earlier melanotropic peptides . NMR spectroscopy and molecular modeling suggest a “reverse-turn” conformation at its active site, which underpins its activity on melanocortin receptors .

Melanotan II acts as a non-selective agonist of melanocortin receptors, binding to MC1R (primarily responsible for melanogenesis), MC3R, and MC4R . Its propensity to improve skin pigmentation has made it popular in cosmetic research, while its modulation of other melanocortin receptors opens doors for studies in sexual function and appetite regulation . This “pharmacologically noisy” molecule delivers potent effects across multiple receptor subtypes, making it a valuable but complex tool for receptor pharmacology research .

The Scientific Foundation of Melanotan II Research

Discovery and Development

Melanotan II was created as a counterpart to the body’s naturally occurring hormone, alpha-melanocyte-stimulating hormone (α-MSH), and was designed to enhance the body’s natural tanning process by activating melanocortin receptors . Early research focused on its capacity to increase melanin formation and provide protection against sun-induced skin damage . However, during clinical investigations, researchers discovered that Melanotan II affected other physiological functions beyond pigmentation, including appetite regulation and sexual arousal, leading to potential applications in obesity and erectile dysfunction research .

Mechanism of Action: Melanocortin Receptor Activation

Melanotan II works by mimicking the effects of α-MSH on the melanocortin receptor system, which comprises five receptor subtypes (MC1R to MC5R) that control a range of physiological functions including pigmentation, appetite, and sexual function .

MC1R Activation and Melanogenesis: Of these receptors, MC1R is principally responsible for regulating melanogenesis—the process that produces skin pigmentation. When Melanotan II binds to MC1R on melanocytes, it triggers a signaling cascade that pushes the melanocyte toward producing more eumelanin (brown-black pigment) over pheomelanin (red-yellow pigment) and increases overall melanin content . This results in a darker, more UV-resistant skin phenotype over time—similar to how some individuals naturally tan more deeply due to their MC1R variants, but induced pharmacologically .

MC3R and MC4R Activation: Melanotan II’s broader receptor affinity extends its pharmacology well beyond the skin. By acting on MC3R and MC4R in the central nervous system, it modulates appetite, energy homeostasis, and sexual function . This explains why researchers observe effects such as appetite suppression, changes in libido, and erectile responses in experimental models . The cyclic design of Melanotan II improves stability and potency, so lower doses still exert strong effects across multiple receptor systems .

Pharmacokinetic Profile

Melanotan II is typically administered subcutaneously, where it is absorbed into the bloodstream, distributed through the body, and then cleared by enzymatic degradation and renal excretion . Available data from early-stage studies suggest a plasma half-life on the order of about 30 minutes to a couple of hours, with many sources clustering around approximately 1 hour . This short plasma half-life is typical for unmodified peptides that are not pegylated, lipidated, or otherwise engineered for extended circulation.

However, a short plasma half-life does not mean a short functional effect. Once Melanotan II binds melanocortin receptors in melanocytes and downstream signaling initiates, the biological cascade—upregulated melanin synthesis, packaging in melanosomes, and transfer to keratinocytes—continues well after the peptide level falls . This explains why a brief spike in blood levels can translate into pigmentation effects over several days or weeks.

Key Research Applications

Melanogenesis and Pigmentation Research

Melanotan II is extensively used in studies investigating skin pigmentation mechanisms and melanin synthesis pathways. Early Phase I clinical studies demonstrated that subcutaneous injections of Melanotan II produced increased pigmentation in the face, upper body, and buttock, as measured by quantitative reflectance and visual perception just one week after dosing ended . These results confirmed that MT-II has tanning activity in humans given only 5 low doses every other day by subcutaneous injection .

Research has shown that Melanotan II can induce significant changes in skin color in vivo, such as transforming the pelage color of mice from light yellow to dark brown or black upon subcutaneous administration, demonstrating its potent melanogenic efficacy . The peptide also enhances tyrosinase activity—a key enzyme in melanin synthesis—for days after initial exposure, suggesting a lasting effect at the transcriptional or translational level .

Erectile Dysfunction and Sexual Function Research

During early clinical trials, researchers observed a notable effect on sexual function. A stretching and yawning complex appeared to correlate with the onset of spontaneous, penile erections which were intermittently experienced for 1–5 hours after MT-II dosing, depending on the dose administered . This finding prompted research into the possibility of using Melanotan II to treat sexual dysfunction, particularly erectile dysfunction, where it could offer an alternative to conventional treatments .

The recommended single MT-II dose for erectile dysfunction research, based on Phase I studies, is 0.025 mg/kg/day . Studies on how Melanotan II affects sexual function have revealed that it may heighten sexual desire and improve overall sexual satisfaction, making it relevant for broader research into sexual health and dysfunction .

Appetite Regulation and Obesity Research

Melanotan II’s role in regulating appetite and energy balance represents another intriguing research application. By acting on MC4R to regulate energy balance and decrease food intake, Melanotan II has potential as a research tool for studying obesity and metabolic disorders . The peptide’s ability to affect hunger presents a novel avenue for investigating metabolic pathways and potential therapeutic interventions.

Photoprotection and Skin Cancer Research

Melanotan II was initially investigated for its potential to prevent skin cancer by increasing melanin formation and providing protection against damaging UV radiation . This is especially relevant for fair-skinned individuals who are more susceptible to melanoma. By boosting the body’s natural defenses against UV radiation through increased melanin production, Melanotan II may help reduce the chance of sunburn and subsequent skin damage . Research in this area continues to explore the peptide’s potential role in photoprotection strategies.

Melanocortin Receptor Pharmacology

Melanotan II serves as a valuable research tool for studying melanocortin receptor pharmacology and signaling pathways. Its non-selective agonist activity across MC1R, MC3R, and MC4R allows researchers to investigate receptor interactions, signal transduction mechanisms, and downstream effects in various tissue types .
